离子交联法制备叶酸-壳聚糖微球工艺研究 被引量:7

摘要 目的:制备叶酸-壳聚糖微球。方法:利用壳聚糖在酸性条件下质子化的氨基与叶酸在碱性条件下去质子化的羧基间静电相互作用,通过离子交联法制备叶酸-壳聚糖微球。利用动态光散射、扫描电镜、以及红外、紫外等技术对微球的结构、平均粒径、粒径分布、形态特征进行了研究。并考察了叶酸与壳聚糖浓度、叶酸与壳聚糖的质量比、滴加速度、反应温度及搅拌速度等因素对微球平均粒径的影响。结果:所得微球平均粒径范围很大,为18-90um,粒径分散指数为1.10±0.02。扫描电镜下观察其形态圆整,大小均匀,无粘连。在叶酸浓度为0.5mg/ml,壳聚糖浓度为2mg/ml;叶酸与壳聚糖质量比为0.25;滴加速度为1滴/秒;反应温度为55℃;搅拌速度为900-1000r/min的条件下,所制备的叶酸壳聚糖微球粒径最小,平均粒径为18.07um。结论:该方法可成功制备叶酸-壳聚糖微球。
