摘要
目的研究槲皮素对Cu2+诱导的β淀粉样蛋白片段(Aβ1~40)聚集和细胞毒性的影响。方法采用硫磺素T荧光标记研究槲皮素对Cu2+诱导的Aβ1~40聚集的影响,利用MTT法测定细胞活力。结果槲皮素可有效抑制Cu2+诱导的Aβ1~40聚集,对已形成的Cu2+-Aβ1~40聚集物有明显的解聚作用,能有效防止Cu2+和Aβ1~40共同诱导的SH-SY5Y细胞活力下降。结论槲皮素对老年痴呆具有潜在的防治作用。
Objective To investigate the effects of que rcetin on the aggregation of Aβ1~40 induced by Cu2+ and on the neurotoxicity of Cu2+-A β1~40 aggregation. Methods The aggregation of Aβ1~40 induced by Cu 2+ was detected by Thioflavin T fluorescence spectroscopy.The cell viability was evaluated by MTT assay. Results Quercetin could inhibit Cu2+-induced aggre gation of Aβ1~40 and the preform ed Cu2+-Aβ1~40 aggregations could be solubilized by quercetin.Th e neurotoxicit y of Cu2+-Aβ1~40 aggregation to SH-SY5Y cells was attenuated by q uercetin and the cell viability loss was significantly decreased. Conclusions Quercetin coul d be as neuroprotective agent for the treatment of Alzheimer′s disease.
