摘要
目的研究二氢杨梅素(DMY)对Cu2+诱导的β淀粉样蛋白片段(Aβ1-40)聚集和细胞毒性的影响。方法采用硫磺素T荧光标记研究DMY对Cu2+诱导Aβ1-40聚集的影响,MTT法检测SH-SY5Y细胞活力的变化。结果 DMY能明显抑制Cu2+诱导的Aβ1-40聚集,对已形成的Cu2+-Aβ1-40聚集物有解聚作用,且能有效防止Cu2+和Aβ1-40共同诱导的SH-SY5Y细胞活力的下降。结论 DMY对阿尔茨海默病具有潜在的防治作用。
Objective To investigate the effects of dihydromyricet(DMY) on the aggregation of β-amyloid (Aβ1-40) induced by Cu2+ and the neurotoxicity of Cu2+-Aβ1-40 aggregates. Methods The aggregation of Aβ1- 40 induced by Cu2+ was detected by thioflavin T fluorescence spectroscopy, and the change of SH SY5Y cell viability was evaluated by MTT assay. Results DMY significantly inhibited Cu^2+-induced aggregation of Aβ1-40 and depolymerized the preformed Cu^2+-Aβ1-40 aggregates. Moreover,DMY could effectively prevent the decreasement of cell viability induced by Cu2+ and Aβ1-40. Conclusion DMY may be taken as a neuroprotective agent for the treatment of Alzheimer's disease.
