Determination of vitamin C in milk powder by high performance liquid chromatography

摘要 目的:建立高效液相色谱法测定乳粉中维生素C的含量的分析方法。方法:样品用3%偏磷酸溶液提取,正己烷脱脂净化,用高效液相色谱仪(C18柱)分析,流动相为0.05 mol/L磷酸二氢钾(pH=2.5),流速1.0 ml/min,二极管阵列检测器紫外检测波长为240 nm,维生素C以光谱图和保留时间定性,外标法峰面积定量。结果:乳粉样品在5 mg/kg~100 mg/kg添加水平范围内,回收率在84.6%~108%之间,相对标准偏差3.96%~7.81%,方法的定量限为5 mg/kg。结论:该法实用、准确,适用于乳粉中维生素C检测。 Objective:To develop a high performance liquid chromatographic(HPLC) method for the determination of vitamin C in milk powder.Methods: Sample was extracted with 3% metaphosphate acid solution,and the extract was then treated with n-hexane for the removal of lipids prior to HPLC analysis using a mobile phase of 0.05 mol/L potassium dihydrogen phosphate solution(pH=2.5) at a flow rate of 1.0 ml/min with PDA detection at the 240 nm.Vitamine C was affirmed according to PDA spectrogram and retention time and quantified by peak area with external standard method.Results: The recoveries of the method spiked at 3 levels(5 mg/kg,50 mg/kg and 100 mg/kg) were within the range of 84.6%~108%,with RSDs ranging from 3.96% to 7.81%.The detection limit of the method was 5 mg/kg.Conclusion: The method is practical,accurate and suitable for the determination of vitamin C in milk powder.
