摘要
采用液体摇瓶培养法,对草菇菌(Volvariella volvacea) 产超氧物歧化酶(SOD) 的培养条件进行初步研究。结果表明:草菇菌丝最适的培养基为:麦麦夫3 % ,蔗糖2 % ,KH2PO40-1 % ,MgSO4·7H2O0 .05 % ,培养条件为:温度35 ℃,pH7 .5 ,摇瓶转速100 r/min 和培养周期48 ~56 h。每克鲜草菇菌丝可得SOD186 单位。聚丙烯凝胶电泳表明,该菌丝的SOD 同工酶谱为单一条带。
Initial studies on liquid culture and determination of superoxide dismutase of V·volvacea were reported.The results showed that the perferable medium for the hypha growth was listed as follows:bran 3%,sucrose 2%,potassium dihydrogen phosphat 1‰,magnesium sulfacte 0.5‰.The culture conditions of temperature,pH,rotation rate,and culture period were 35℃,7.5,100 r/min and 48~56 h,respectively.The determination indicated that SOD activity is 186 units in per-gram wet hypha.PAGE of isozyme analysis is only one band.
