摘要
对一株来自东祁连山高寒草地土壤具有纤维素分解能力的真菌的培养特性进行了研究。结果表明:供试菌株的菌丝生长温度5~35℃,最适生长温度30℃;在参试的pH范围均能生长,并且在pH7.08的环境生长最快;供试菌株具有一定的耐盐和抗干旱胁迫的能力;对不同的碳、氮源利用能力不同,在供试的11种碳源和12种氮源中,乳糖和大豆蛋白胨分别为最适碳源和氮源。以1.5%的水琼脂培养基为对照菌株,在黑麦草(Lolium perenne)浸提液培养基上生长较快,三叶草(Trifolium repens)的浸提液培养基上的生长速度较慢,其余的天然纤维素浸提液培养基上的生长速度差异不明显。
Cultural characteristic of one cellulose-decomposing stain of fungus from alpine grassland soil in eastern Qilian Mountains were studied.The results showed that this strains was able to grow at 5~35 ℃.The optimal cultural temperature and pH values for mycelial growth was 30 ℃ and 7.08 respectively.The strain has ability of salt and PEG stress tolerance.It varied in its viability on media containing different carbon and nitrogen nutrients.The preferential carbon and nitrogen source was lactose and soybean peptone,respectively.Compare to the 1.5% water agar medium,the growth of the stain was faster in ryegrass extract,but slower in clover extract.The growth of the mycelium was not significantly difference in medium with different natural cellulose extract.
