摘要
目的:观察 C D4 + αβ T 细胞和γδ T 细胞对结核杆菌( Mtb) 不同抗原的增殖反应,以及活动性肺结核患者外周血中这两类 T 细胞亚群的数量和功能的变化。方法:用 Mtb 分泌性蛋白抗原和耐热性低分子多肽抗原刺激健康人和肺结核患者的外周血 T 细胞,用荧光标记特异性单抗在流式细胞仪上检测新鲜分离和培养10 d 后的细胞表型,以计数扩增的 T 细胞亚群数量。结果:活动性肺结核患者外周血中 C D4 + αβ T 细胞的数量比正常对照明显减低,而γδ T 细胞的数量无明显变化; Mtb 分泌性抗原仅能刺激 C D4 + αβ T 细胞增殖,而耐热性多肽抗原能刺激两类 T 细胞亚群增殖;活动性肺结核患者外周 C D4 + αβ T 细胞对 Mtb 分泌性抗原以及γδ T 细胞对 Mtb 耐热性多肽抗原的增殖反应均明显降低。结论: C D4 + αβ T 细胞和γδ T 细胞均参与机体抗 Mtb 感染的保护性免疫机制。
Objective:Investigation of changes in numbers and proliferation responses of CD4 +αβ T cells and γδ T cells in peripheral blood of patients with advanced pulmonary tuberculosis.Methods:Two different Mtb antigen, culture filtrates as secrete protein antigen and heat resistant low molecular peptide antigen were applied to stimulate T cell sub populations of patients and normal subjects and responded cells were phenotyped by flow cytometry.Results:The proportion of CD4 + T cells in peripheral blood of patients with advanced TB was reduced in comparison with normal subjects, yet absolute number of the cells was not significantly changed. Mtb secrete protein antigen was able to stimulate CD4 + T, but not γδ T lymphocytes for proliferation in normal subjects, whereas heat resistant peptide antigen derived from Mtb can stimulate both CD4 + and γδ T cells. In patients with advanced TB, the proliferation response of CD4 + T cells to secrete protein antigen and that of γδ T cells to heat resistant peptide antigen were severely impaired.Conclusions:The results from this paper showed the clinical evidences and supported the hypothesis that both CD4 + T and γδT cells have participated the protective immunity of host against Mtb infection.
