摘要
筛选了真菌、细菌和放线菌共1121株,其中有β-HexNAcase 活力的有237株,在所筛选菌中只要有β-GlcNAcase 活力,也就有β-GalNAcsse 活力,但两酶比值(β-GlcNAcsse/β-Gal-NAcsse)不同。所筛选出的溜曲霉(Asg.tamarii)S215为由土壤中分离的,其产酶最适条件为5%麸皮液体培养基,28—30℃振荡培养5—6天,补充碳源如纤维二糖、葡萄糖胺、半乳糖胺或者补充氮源(NH_4)_2SO_4及 NH_4NO_3可以稍稍提高酶活。在溜曲霉的培养液中除β-GlcNAcase 及β-GslNAcase 外,还有α-半乳糖苷酶、β-半乳糖苷酶、β-葡萄糖苷酶、β-岩藻糖苷酶及α-甘露糖苷酶。
About 1200 strains of microorganismswere screened including fungi,actinomyces,and bacteria,in which 237 strains producingthe enzyme desired.The results showed thatthe β-GlcNAcase and β-GalNAcase alwaysco-existed in one strain,though may be indifferent ratio.From strains mentioned above the authorsscreened out a potent β-N-acetylhexosamini-dase producing strain,Aspergillus tamariiS215,from the soil sample.The optimal con-ditions for enzyme production were as fol-lows:The microorganisms was inoculated ina 5% wheat bran suspension,cultured at 28—30℃ on shaker for 5—6 days.The pro-ductivity can be moderately enhanced by the(?)ddition of cellobiose or glucosamine or ga-lactosamine or by the extra suppliement of(NH_4)_2SO_4 and NH_4NO_2 as N sources.Inthe culture filtrate of Asp.tamarii,the α,(β)-galactosidase,β-glucosidase,α-mannosi-dase and β-fucosidase were also found.
