摘要
目的:对人重组骨形态发生蛋白2(rh B M P2)和 β转化生长因子( T G Fβ)在诱导成骨的协同作用进行探讨. 方法:210 只 B A L B/c 小鼠随机分为 3 组,每组 70 只,试验侧均位于右后肢,设立 rh B M P2/牛松质骨载体、单纯牛松质骨载体为对照组. 分别于术后 12 h ~21 d 共 11 个时间点取材,观察其诱导成骨过程. 结果:rh B M P2/ T G Fβ/牛松质骨载体组在诱导间充质细胞增殖、分化,软骨细胞及新生骨形成方面均早于rh B M P2/牛松质骨载体组,成骨量优于 rh B M P2/牛松质骨载体组,其碱性磷酸酶水平高峰较对照组提前,其组织钙含量明显高于对照组( P< 0.05). 而单纯牛松质骨载体组在 21 d仅出现了少量增殖的间充质细胞. 结论:rh B M P2 和 T G Fβ在诱导成骨调节中存在着协同作用.
AIM: To study a possible synergism between recombinant human bone morphogenetic protein protein 2(rhBMP 2) and transforming growth factor β(TGF β) in osteoinduction and osteogenesis. METHODS: 210 BALB/c mice were divided into three groups at random, 70 in each group, and composites were implanted in the right thigh pouchs. Mice receiving composite of rhBMP 2/ carrier and carrier only served as the controls. In order to investigate the process of osteogenesis, histological analysis was performed 12 hours, 1, 2, 3, 5, 7, 9, 11, 14, 17 and 21 days after the implantation; meanwhile, alkaline phosphatase activity and calcium content were measured 7, 14 and 21 days after the implantation. RESULTS: Proliferation and differentiation of mesenchymal cells, chondrogenesis and osteogenesis induced by the composite of rhBMP 2/TGF β/carrier appeared earlier than those induced by the composite of rhBMP 2/carrier. The amount of osteogenesis was greater than that in the group of rhBMP 2/carrier. Peak of ALP activity in the experimental group occurred earlier than those in the controls and calcium content was higher than those in the controls obviously. However, only a few proliferating mesenchymal cells were observed in the carrier group 21 days after the implantation. CONCLUSION: rhBMP 2 and TGF β exert a synergetic action in regulating osteoinduction and osteogenesis.
