摘要
目的:研究地黄饮子对老年性痴呆神经保护作用及其机制。方法:通过细胞免疫法检测各组PC12细胞ChAT、tau的表达。结果:Aβ25-35损伤模型组PC12细胞中ChAT表达明显低于空白组(P<0.01),而加入地黄饮子脑脊液后可提高ChAT的表达(P<0.05或P<0.01),中药组与安理申组比较无显著性差异(P>0.05);Aβ25-35损伤模型组PC12细胞中tau表达明显高于空白组(P<0.01),而加入地黄饮子脑脊液后,可剂量依赖性降低tau的表达(P<0.01),且中高组、中中组疗效较好,与安理申组比较有显著性差异(P<0.01或P<0.05)。结论:地黄饮子能明显提高Aβ25-35损伤时PC12细胞ChAT的表达,能抑制细胞微管相关蛋白tau的表达,起到神经保护的作用,从而达到防治AD的目的。
Objective:To investigate the neuroprotective effect and mechanism of Di Huang Yin Zi(DHYZ) on AD.Methods:To investigate the expression of ChAT and tau of PC12 in each group by using cellular immunity.Results:1.The expression of ChAT in the group of PC12 induced by Aβ25-35 was significantly lower than that of control group(P0.01),and after adding CSF DHYZ,the expression of ChAT was enhanced(P0.05 or P0.01).There was no difference between Chinese medicine group(CMG) and the Aricept group(P0.05).2.The expression of tau in the group of PC12 induced by Aβ25-35 was significantly heigher than that of control group(P0.01),and after adding CSF DHYZ,the expression of tau was decreased(P0.01) in a dose-dependent pattern.The high-does CMG and the moderate-dose CMG were better than that of other groups,and better than the Aricept group with a singificant difference(P0.01).Conclusion:DHYZ could obviously improve the expression of ChAT of PC12 induced by Aβ25-35,and significantly decease the expression of canaliculus-associated protein-tau of PC12 induced by Aβ25-35.So DHYZ can protect the neurons,prevent and treat AD.
