摘要
人工重组的白细胞介素1β(IL-1β)多次免疫兔和豚鼠,获取高效价的兔抗IL-1β抗体。用氯氨T法制备125I标记IL1β,经Sephadex G-25纯化。该法测定范围0.06—4ng/m l,批内和批间误差分别小于5% 和10% 。正常人血清含量为0.24±0.08ng/m l(n=138)。人粒细胞系HL60(106)细胞体外培养24 小时,不能检出上清液中IL-1β,钙离子载体A23187和磷脂酶A2 抑制P-BPB也不能显著提高细胞释放IL-1β。另外,肝癌甲胎蛋白阳性患者血清中IL-1β水平同正常人没有显著差异。但是,雄性老龄大鼠血清IL-1β水平明显高于雌性老龄大鼠(P< 0.01)
Only the high effect antibody of rabbit IL\|1β was obtained by immunizing rabbits and guinea pigs with recombinant IL\|1β many times. The IL\|1β was iodinated with chloramine\|T methods and purified by the Sephadex\|G25 chromatograph column.The determine range of this method was about 0.06—4ng/ml and the lowest detecting level was 0.06ng/ml.The mean errors in group and between groups were less than 5% and 10% respectively.The level of IL\|1β in normal peoples was 0.24±0.08ng/ml(n=138).The liberation of IL\|1β from HL\|60 cells was not obviously promoted by cultured with calcium ionophore(A 23187 ) and phospholipase A\-2 inhibitor (P\|BPB) for 24 hours in vitro.Otherwise,there were no differences in serum IL\|1β level between normal patients and liver cancer patients who was positive in serum alphafetoprotein.Whereas,the IL\|1β in serum of aged male rats was significantly higher than that of aged female rats(P<0.01).\;
