摘要
目的从人心肌组织中提取和纯化心肌特异性肌钙蛋白，为建立心肌梗塞病人免疫诊断试剂准备抗原。方法取新鲜人心室肌组织经捣碎离心提取，ＤＥＡＥ５２柱层析纯化，ＳＤＳ－ＰＡＧＥ凝胶电泳鉴定。结果１００ｇ心室肌可提取３７ｍｇ纯品肌钙蛋白。结论本次提取的心肌特异性肌钙蛋白纯度较高，能作抗原使用。
Objective To purify some cardiac troponin T(cTnT)
from human heart as the antigen for developing the immune diagnosis method of myocardial
infarction.Methods Human heart tissue obtained less than two hours post mortem was
homogenized and centrifuged.The DEAE 52 cellulose column chromatography and
SDS-polyacrylamide gel electrophoresis were employed for purification and identification of
cTnT.Results 37 mg cTnT was obtained from 100g left ventricular muscle of human cardiac.
Conclusion The purity of cTnT purifed in this study is higher ,and it can be used as antigen
