摘要
目的 制备抗人心肌肌钙蛋白T的单克隆抗体。方法 利用纯化的人心肌肌钙蛋白T免疫BALB/C小鼠 ,常规方法制备抗人心肌肌钙蛋白的单克隆抗体 2株N15C和N16D。结果 利用普通ELISA方法检测 2株单抗 ,小鼠腹水N16D滴度可达 1/ 32 0 0 0 ,N15C滴度达 1/ 80 0 0 ;斑点酶联免疫吸附试验证明N16D对急性心肌梗塞患者血清中出现的肌钙蛋白T有较好的亲和性。
Objective To prepare the monoclonal antibody(McAb) to human cardiac troponin T(CTnT).Method The two specific McAb to human cardiac troponin T,N 15C and N 16D,were routinely developed by immunizing the BALB/C mice with purified CTnT.Result The titers of anti-CTnT McAb,N 15C and N 16D,purified from ascetic fluid of mouse respectively reached to 1/8000 and 1/32000 by ELISA measured;The McAb anti-CTnT N 16D was capable of specific binding CTnT in serum from patients with acute myocardial infarction by dot-ELISA.Conclusion The anti-troponin T McAb N 16D possesses affinity and specificity to human troponin T.
