摘要
目的:建立测定紫草中β,β′-二甲基丙烯酰阿卡宁含量的方法。方法:采用改进后的高效液相色谱法,以流动相制备供试品溶液。色谱柱为Kromasil100-5C18(150mm×4.6mm,5μm),流动相为乙腈-水-甲酸(700∶300∶0.5),流速为1.0mL·min-1,检测波长为275nm。结果:β,β′-二甲基丙烯酰阿卡宁的进样量在0.16~3.20μg范围内与峰面积积分值呈良好线性关系(r=0.9999);平均回收率为99.46%,RSD=1.90%(n=6)。结论:该方法简便、易行,方法验证符合要求,可用于紫草的含量测定。
OBJECTIVE:To establish a method for the content determination of β,β'-dimethylacrylalkannin in Radix Arnebiae.METHODS:Improved HPLC method was adopted and samples were prepared by mobile phase.Kromasil 100-5 C18(150 mm×4.6 mm,5 μm) column was used with mobile phase consisted of acetonitrile-water-formic acid(700:300:0.5)at flow rate of 1.0 mL· min-1.The detection wavelength was set at 275 nm.RESULTS:β,β'-dimethylacrylalkannin had good linearity in the range of 0.16-3.20 μg(r=0.999 9) with an average recovery of 99.46%(RSD=1.90%,n=6).CONCLUSION:The method is simple and feasible for content determination of Radix Arnebiae.Method verification is up to the standard.
