摘要
目的:建立清肺抑火丸中苦参碱和氧化苦参碱的含量测定方法。方法:采用高效液相色谱法。色谱柱为Inertsil NH2柱,流动相为乙腈-无水乙醇-3%磷酸溶液(80∶10∶10),检测波长为220 nm,流速为1.0 mL·min-1,柱温为25℃,进样量为10μL。结果:苦参碱浓度在10.0~50.0μg·mL-1范围内与峰面积积分值线性关系良好(r=0.999 5),氧化苦参碱浓度在28.0~84.0μg·mL-1范围内与峰面积积分值线性关系良好(r=0.999 5)。苦参碱和氧化苦参碱的平均加样回收率分别为99.0%(RSD=2.58%,n=6)和101.0%(RSD=2.95%,n=6)。结论:本法简便快捷、结果准确,可用于测定清肺抑火丸中苦参和氧化苦参碱的含量。
To establish a method for the content determination of matrine and oxymatrine in Qingfei yihuo pills. METHODS: HPLC method was adopted. The determination was performed on a Inertsil NH2 cohmm using acetonitrile-absolute ethyl alcohol -3% phosphoric acid (80: 10: 10) as mobile phase. The flow rate was 1.0 mL·min^-1 and the detection wavelength was 220 nm. Column temperature was 25 ℃. The injection volumn was 10 μL. RESULTS: The linear range of matrine was 10.0-50.0 μg·mL^-1(r=0.999 5) and that of oxymatrine was 28.0-84.0μg·mL^-1 (r=0.999 5). The average recoveries of matrine was 99.0% (RSD=2.58%, n=6) and oxymatrine was 101.0% (RSD=2.95%, n=6). CONCLUSION: The method is simple, rapid and accurate, and it can be used for the content determination of matrine and oxymatrine in Qingfei yihuo pills.
