摘要
目的:建立同时鉴别苦参药材8种生物碱并测定5种生物碱含量的高效液相色谱法。方法:采用 Diamonsil^(TM) C_(18)柱(5μm,250 mm×4.6 mm),以0.05 mol·L^(-1)磷酸二氢钠水溶液-乙腈-甲醇-高氯酸钠(940 mL:50 mL:10 mL:10 g,磷酸调节 pH 4.4)为流动相,流速1 mL·min^(-1),检测波长210 nm,柱温0~29 min,40 ℃;30~39 min,50℃;40~45 min,40℃。结果:不同药材市场的苦参所含生物碱成分的种类和含量均有差异。结论:本法灵敏、准确、专属性好,适用于苦参药材中生物碱成分的鉴别和含量测定。
Objective: To establish an HPLC method to identify the eight alkaloids and determine the five of them in Sophoraflavescens Ait. simultaneously. Method: The method was conducted on a Diamonsil^TM C18 column (5 μm, 250 mm×4.6 mm) with 0.05 mol·L^-1 sodium dihydrogen phosphate - acetonitrile - methyl alcohol - sodium perchlorate (940 mL:50 mL:10 mL:10 g, pH 4.4) as mobile phase, flow rate 1 mL·min^-1, detecting at 210 nm and column temperature at 0 - 29 min, 40℃; 30 - 39 min, 50℃; 40 - 45 min, 40℃. Results: The components and quantity of the alkaloids in Sophora flavescens Ait. from different herbs markets are different. Conclusion: The method is sensitive, accurate and specific for the identification and determination of the alkaloids in Sophora flavescens Ait.
