紫杉醇改变FADD在食管鳞癌中的表达 被引量:2

摘要 目的研究紫杉醇对人食管鳞癌细胞株Eca-109中Fas相关死亡结构域蛋白(Fas associat-ed death domain protein,FADD)表达的影响,探讨紫杉醇在食管鳞癌治疗中的作用机制。方法流式细胞术检测不同浓度紫杉醇处理下,人食管癌细胞株Eca-109细胞凋亡率,同时采用Western blotting分析细胞中FADD的表达变化。结果紫杉醇诱导人食管癌细胞株Eca-109细胞凋亡呈剂量依赖性;随着紫杉醇剂量的增加,实验组Eca-109细胞中FADD表达呈显著增高。结论一定剂量的紫杉醇可以提高FADD在食管癌细胞中的表达、促进食管癌细胞的凋亡,这可能是紫杉醇治疗食管癌的分子机制之一。 Objective To observe the changes of FADD expression in esophageal carcinoma cell 109(Eca109)induce by Paclitaxel.Methods With different concentration of paclitaxel, the apoptosis rates of Eca109 cells and protein expression levels of FADD in Eca109 cells were detected. Results The apoptosis rates of Eca109 cells were increased with concentration of paclitaxel rising,and protein expression levels of FADD in Eca109 cells were increase simultaneously. Conclusion Paclitaxel induced Eca109 cell apoptosis via changing protein expression levels of FADD.
