摘要
目的 :观察不同浓度的紫杉醇对食管癌细胞 Eca10 9的影响。方法 :应用四甲基偶氮唑蓝 (MTT)实验、细胞形态学观察及流式细胞仪等方法对人食管癌细胞 Eca10 9进行检测和观察。结果 :MTT实验显示紫杉醇可抑制食管癌细胞增殖 ;光镜及电镜可发现药物作用组细胞核固缩、解聚以及凋亡小体 ,流式细胞仪检测显示 G1 峰前有明显的凋亡峰。细胞周期分析提示紫杉醇可将 Eca10 9细胞阻滞于 G2 - M期 ,且与浓度相关。结论 :紫杉醇对人食管癌细胞系 Eca10 9细胞的生长有明显的抑制作用 ,使细胞分裂阻滞于 G2 - M期 ,并诱导肿瘤细胞凋亡。
Objective:To observe the effects of paclitaxel on esophageal carcinoma cell 109(Eca109) with different concentration and duration. Methods: With different concentration and duration of paclitaxel, the Eca109 cell activity, morphologic changes and cell DNA distribution were studied. Results:Paclitaxel could inhibit the proliferation of Eca109 cells in vitro, which was associated with arrest of G 2 M phase and with inducing apoptosis. Chromation condensation, nucleic fragmentation and apoptic body formation were found; folw cytometry analysis showed a sub G 1 cell peak. Conclusion:Paclitaxel can induce apoptosis and block cell cycle of esophageal cells.
