摘要
研究了 5-甲基 - DL-色氨酸 ( 5- MT)、丙二酸钠等 11种代谢抑制剂对红豆杉悬浮培养细胞生长和紫杉醇生物合成速率的影响 ,筛选到 5- MT等 3种对紫杉醇生物合成有促进作用的抑制剂。抑制剂在稳定期添加对红豆杉细胞生长无强烈的抑制作用 ,更有利于紫杉醇的生物合成。各抑制剂与前体和真菌诱导子协同作用能提高增产效果 ,增产大小依次为 5- MT、马来酸和丙二酸钠。其中 50 mg· L-15- MT使紫杉醇含量提高近10倍 ,10 mg· L-1马来酸和 10 mg· L-1丙二酸钠都增产约 3倍。
The effects of 11 inhibitors such as 5 methyl DL tryptophan(5 MT)on cell growth and taxol yield in Taxus chinensis suspension cultures were studied in this paper.Three inhibitors i.e.5 MT,sodium malonate,maleic acid that can improve taxol yield have been screened,when added at stationary phase these inhibitors not only increase taxol yield,but also do less harm to cell growth of yew suspension cultures.When combined with procursors and fungal elicitors the yield improving abilities of all inhibitors were enlarged.Especially taxol yield was 10 fold increased when 5 MT was added at a terminal concentration of 50 mg·L -1 combined with procursors and fungal elicitors.
