摘要
目的:确定杜仲抗皮肤光老化的有效部位。方法:杜仲乙醇提取物经大孔树脂制备不同浓度乙醇梯度洗脱部位,分别作用于HaCaT细胞光老化模型,以细胞形态、细胞活性及细胞培养基中LDH、MDA、SOD活力为评价指标,筛选并确定杜仲抗皮肤光老化的有效部位。结果:与模型组相比,浓度为125μg.mL-1的50%乙醇大孔树脂洗脱部位能提高细胞活性(P<0.05),使培养基中LDH降低(P<0.01)、MDA活力降低(P<0.05)、SOD活力提高(P<0.05);62.5μg.mL-1的给药浓度能降低LDH活力(P<0.05)。结论:50%乙醇大孔树脂洗脱部位能清除氧自由基而提高SOD活力、降低MDA活力,保护细胞膜免受损伤,降低LDH的泄露,具有一定的抗皮肤光老化作用。
Objective : To ensure the effective part of Eucommia for anti - photoaging. Methods : Ethanol extract of Eucommia macroporous resin prepared by different parts of the concentration gradients of ethanol. The cell model of photoaging was acted with elution positions. The effective portion of anti - photoaging of eucommia was ensured by the index of cell morphology, cytoactive and LDH, MDA, SOD activity. Results : At the dosage of 125 2μg·mL-1, cytoactive were raised( P 〈 0. 05 ) ;LDH activity were decreased (P 〈 0.01 ) ; MDA activity were decreased ( P 〈 0. 05 ) ; SOD activity wasincreased (P 〈 0. 05 ) when 50% elution site were added. Conclusion : Oxygen free radicals were protected membranes from ethanol eluting could resist scavenged; SOD activity were increased and MDA activity were decreased, and it damage and reduce the leakage of LDH.
