摘要
采集了129份屠宰鸡血清样品,采用酶联免疫吸附试验检测了鸡血清样品中HBsAg、HBeAg、HBsAb、HBcAb、HBeAb等乙型肝炎诊断指标;用免疫电镜方法观察了血清样品中乙型肝炎病毒样粒子。采用免疫组织化学技术检测了肝中乙型肝炎病毒相关抗原的分布,并对鸡肝的S基因进行了PCR检测及同源性比对。结果显示,HBsAg、HBeAg、HBsAb、HBcAb、HBeAb的血清阳性率分别为28.68%、53.49%、17.05%、4.65%和9.3%,其中3份血清同时呈HBsAg、HBeAg阳性,阳性率为2.33%。电镜负染色样品观察可见大量密集排列的病毒样粒子,形态和大小类似于人乙型肝炎病毒Dane颗粒和小球状颗粒。鸡肝中HBsAg、HBcAg免疫组织化学检测结果显示,阳性率为39.06%~62.07%。应用PCR技术扩增出的2株乙型肝炎病毒的核酸序列片段与人乙型肝炎病毒的同源性分别高达92.2%和97.9%。以上结果说明,部分屠宰鸡可能存在乙型肝炎病毒感染。
A total of 129 sera samples and 193 liver samples were collected from the slaughtered chic-kens for a serological survey.HBsAg,HBsAb,HBcAb,HBeAg and HBeAb were examined by ELISA.The virus-like particles were examined by transmission electron microscope(TEM) .The distribution of relative antigens in the liver of slaughtered chicken was studied by immunohistochemistry.S gene sequences were amplified by PCR from the liver and the comparison of homology was carried out with the S gene sequences.HBsAg,HBsAb,HBcAb,HBeAg and HBeAb seropositive rate were 28.68%,53.49%,1705%,4.65% and 9.3%,respectively.Among all of them,3 sera samples were co-positive for HBsAg and HBeAg.Lots of HBV(hepatitis B virus) -like particles were found in the negative staining serum samples under TEM.Immunohistochemistry detection showed that HBsAg and HBcAg positive liver rates were between 39.06% and 62.07%.Among all of the livers,two strains of HBV were identified.The two detected HBV sequences shared 100% nucleotide sequence identity each other,and 92.2% and 97.9% with the other two human HBV strains.The results suggested that some slaughtered chickens were infected with HBV.
