摘要
[目的]寻找铁线蕨孢子组织培养的适宜培养基,提高其繁殖效率。[方法]以铁线蕨附带成熟孢子的叶片为外植体进行组织培养,找出合适的萌发培养基、诱导培养基、增殖培养基、分化成苗培养基。[结果]铁线蕨孢子的最佳期萌发培养基为1/2MS+活性炭0.5g/L,原叶体在培养基1/2MS+BA2.5mg/L+IBA0.1mg/L上可诱导出GGB,GGB在培养基1/2MS+BA1.5mg/L+IBA0.1mg/L上可不断增殖,在培养基1/2MS+BA0.3mg/L+IBA0.1mg/L上可以分化成苗,同时进行驯化移栽。[结论]该研究为铁线蕨快速繁殖提供了更有效的途径,并为生产企业的大规模种苗生产提供强有力的技术支持。
[Objective] The aim was to find the optimum medium for the tissue culture of Adiantum capillus-veneris conidia and to improve its propagation rate.[Method]With Adiantum capillus-veneris mature conidia on leaf blade as explants,the tissue culture was carried out to find out the optimum germinating medium,inducing medium,multiplication medium and plantlets medium.[Result] Adiantum capillus-veneris conidia germination medium was the best of 1/2MS + activated carbon 0.5 mg/L.Prothallus in the medium 1/2MS + BA 2.5 mg/L+ IBA 0.1 mg/L could induce GGB.GGB in the medium 1/2MS + BA 1.5 mg/L+ IBA 0.1 mg/L could continue to proliferate.In the medium 1/2MS + BA 0.3 mg/L + IBA 0.1 mg/L could differentiate GGB into seedlings,transplanting acclimation.[Conclusion] The rapid propagation of Adiantum capillus-veneri study provides a more effective way,and provides strong technical support for the production of large-scale seed production enterprises.
