摘要 目的评估贝伐单抗(bevacizumab,商品名Avastin)对大鼠角膜上皮缺损愈合速率及对正常大鼠角膜形态结构的影响。方法健康雌性SD大鼠随机分为缺损组和正常组,缺损组包括实验A组和对照A组,均建立角膜上皮缺损模型,每组4只(8眼);正常组包括实验B组和对照B组,每组2只(4眼)。上述实验组双眼均滴5g.L-1的Avastin,对照组均滴生理盐水。A组于造模后12h、18h、24h、36h荧光素染色裂隙灯观察角膜上皮缺损修复情况,直至上皮缺损愈合;B组在连续滴眼7d后做角膜组织病理学观察和透射电镜检查。结果裂隙灯观察显示,在24h内实验A组和对照A组角膜上皮愈合速率一致,36h时角膜上皮均已愈合;实验A组和对照A组在12h、18h、24h角膜上皮缺损平均愈合速率比较,差异均无统计学意义(均为P>0.05);正常组滴眼7d后,与对照B组比较,实验B组的光镜和电镜检查未见角膜结构改变及炎性细胞浸润。结论短期局部应用5g.L-1Avastin对大鼠角膜上皮缺损愈合速率和正常角膜结构无影响。 Objective To evaluate the effect of bevacizumab(Avastin)on healing rate of corneal epithelial defect and corneal morphological structure in normal rats.Methods Healthy female SD rats were randomly divided into 2 groups,the defect group and control group.The corneal epithelial defect models were made in defect group,which was subdivided into experimental group A and control group A,4 rats(8 eyes)in each group.Rats in control group were subdivided into experimental group B and control group B,2 rats(4 eyes)in each group.The 5 g·L-1 Avastin eye drop was given to both eyes of rats in experimental groups,and normal sodium was given to rats in control groups.Healing of corneal epithelial defect was observed by fluorescence staining under slit lamp at 12 hours,18 hours,24 hours and 36 hours after modeling in group A until complete healing of epithelial defect.After continuously treating with eye dropping for 7 days,rats in group B were taken corneal histopathologic observation and transmission light microscope examination.Results Slit lamp observation showed that corneal healing rate was consistent in experimental group A and control group A during 24 hours,the corneal epithelium all healed at 36 hours.The healing rate of corneal epithelial defect showed no significant difference between experimental group A and control group A at 12 hours,18 hours and 24 hours(all P0.05).After eye dropping for 7 days in rats of control group,the corneal histological structure and ultramicrostructure were normal without the inflammatory cell infiltration in all corneas of experimental group B compared with control group B.Conclusion There was no effect on the healing rate of rat corneal epithelium and corneal structure after short-term ocular application of 5 g·L-1 Avastin.
