摘要
目的:通过观察王氏保赤丸与抗生素联合用药对肠道菌群失调小鼠肠黏膜的影响,探讨该药是否能预防和纠正抗生素所致肠道菌群失调后肠黏膜损伤现象,为扩大其适用范围提供实验依据。方法:建立抗生素引起菌群失调小鼠模型对照组,正常对照组,抗生素加高、中、低剂量王氏保赤丸灌胃小鼠模型组,抗生素加妈咪爱西药对照组。与正常小鼠、模型对照组及西药对照组比较,评价王氏保赤丸对小鼠肠道黏膜组织的影响情况。结果:灌胃14天后抗生素加王氏保赤丸高、中、低剂量组和妈咪爱组均未出现模型组的小肠绒毛充血、变平、变短、萎缩、倒伏等现象。与模型组对比,模型加低剂量王氏保赤丸组肠黏膜缺损减轻,小肠绒毛变长,杯状细胞增生;中剂量组病变介于低剂量组和高剂量组之间;高剂量组与妈咪爱组结果类似。回盲部的病理结果与小肠病理结果类似。肝脏病理结果显示除正常组外,各组均对肝细胞有损害,王氏保赤丸各组、妈咪爱组与模型组相比无明显差异。结论:王氏保赤丸对抗生素致菌群失调肠道黏膜有保护和修复作用。高剂量王氏保赤丸组对肠道黏膜保护和修复作用与妈咪爱组相同。二药对抗生素所致肝损害无明显修复作用。
Objective:To observe the change of WanShiBaoChiWan(WSBCW) in the intestine and liver of mice after given lincomycin,and provide data and evidence for proper use of it in clinic.Methods:Antibiotic-induced intestinal dys-bacteriosis mice model group,WSBCW groups of various dose and Western drug control group made to compare the mor-phology of their intestine and liver between groups and with normal mice.The mice were given gastric lavage for 14 days by Lincomycin(0.4mL/d) and high dosage,middle dosage,low dosage WSBCW [147 mg/(kg.d),49 mg/(kg.d),17 mg/(kg.d) ]in WSBCW groups,and Lincomycin(0.4mL/d) in model group,Lincomycin(0.4mL/d) and Medilac-Vita(0.4mL/d) in West drug control group,N.S(0.4mL/d) in normal group.The structure of appendix and small intestine,liver were examined after 14 days.Results:The congestion,flattening,shortening,atrophy,and lodging of small intestinal villosity microvessels of mice that showed in model group were not showed in other groups.The damages of intestinal mucosa were relatively decreased in all dose WSBCW groups compared to model group.The lengthe-ning of small intestinal villosity microvessels,and increasing of goblet cells in high dosage WSBCW group were same with Medilac-Vita group.The observation showed that the changes of ileocecal junction were same with small intestine.Liver cell damage were found in all groups except normal group.Conclusion:WSBCW has protective effect to intestinal tract and decrease the damage of intestinal mucosa caused by antibiotics.
