摘要
目的:建立以反相高效液相色谱法同时测定射干合剂中黄芩苷等5种黄酮类化合物含量的方法。方法:样品经超声提取后,用InertsilODS-3色谱柱分离样品,流动相为乙腈-1‰磷酸(pH2.25,梯度洗脱),流速为1mL·min-1,柱温为室温,检测波长为260nm;外标法定量。结果:黄芩苷、黄芩素、汉黄芩素、射干苷、次野鸢尾黄素的线性范围均为0.05~50μg·mL-1(r均≥0.9989);平均加样回收率为96.52%~101.22%,RSD均≤4.4%(n=9)。结论:本方法简便、准确、灵敏度高,可用于射干合剂中黄酮类化合物多指标成分的含量测定,也可用于其制剂及体内指标成分的研究。
OBJECTIVE:To establish RP-HPLC method for the content determination of 5 kinds of multiple indicator constituents of flavonoids in Belamcanda chinensis mixture such as baicalin.METHODS:Samples were extracted with selective ultrasound extraction.The separation was performed on InertsilODS-3 column with mobile phase consisted of acetonitrile-1‰ phosphoric acid(pH=2.25,gradient elution) at flow rate of 1 mL·min^-1.The column temperature was room temperature and detection wavelength was set at 260 nm.The method was qualified by an external standard mode.RESULTS:The linear ranges of baicalin,baicalein,wogonin,tectoridin and irisflorentin were all 0.05~50 μg·mL-1(r≥0.998 9) with average recoveries of 96.52%~101.22%(RSD≤4.4%,n=9).CONCLUSION:This method is simple,convenient,accurate and sensitive for content determination of multiple indicator constituents of flavonoids in B.chinensis mixture and constituent study of preparation and internal index constituent.
