氟铝联合对体外培养人胚成骨细胞毒作用的形态学观察

摘要 观察氟铝联合对体外培养人胚成骨细胞毒作用。方法:采用无血清体外细胞培养方法,将成骨细胞分成对照组、低铝组、高铝组、低铝低氟组、高铝低氟组及高铝高氟组共6组,以光镜和电镜观察其形态学变化。结果:光镜观察,高铝组和高铝加氟组细胞存活数均明显低于对照组、低铝组和低铝低氟组( P< 0. 05)。高铝组细胞变性坏死率明显高于对照组、低铝组和低铝低氟组(P<0.05)。在高铝组加入不同剂量的氟可明显降低细胞变性坏死率,但仍明显高于对照组、低铝组及低铝低氟组( P< 0. 05),而明显低于高铝组( P< 0. 05);电镜观察,高铝组和高铝加氟组细胞核膜迂曲肿胀,线粒体膜结构不清楚,胞质内含有大量次级和终末溶酶体,多数为髓鞘样结构,而低铝组和低铝低氟组变化轻微。 To study the toxicity of aluminum combined with fluoride on human osteoblasts in culture in vitro. Methods: Using the serum-free medium cell culture in vitro, the cultured osteoblasts were divided into six groups : control , low dose aluminum, high dose aluminum, low dose aluminum-fluoride, low dose aluminum and high dose fluoride, high dose aluxninum fluoride groups. Their morphological changes were observed by light and electron microscope . Results: The observation with light microscope showed that the amount of survival cells in high dose Al and high dose Al-F groups were lower than those in the control, low dose Al and low dose Al-F groups(P < 0. 05 ). The detective rate of cell degeneration and necrosis in high dose Al and high dose Al-F groups were more than those in the control,low dose Al and low dose Al-F groups(P<0 .05). The rates were reduced by adding different dose fluoride to high dose Al group,but were still higher than those in the control,low dose Al and low dose Al-F groups(P<0 .05),and were lower than that in high dose Al group(p<0.05). The observation with electron microcope showed that the nucleic membranes in high dose Al and high dose Al-F groups were tortuous and swollen slightly and the bilayer structure of mitochondria membranes were not clear. There were a large number of secondary and ultimate lysosomes .The majority of them were myelin-like structures. But the changes in low dose Al and low dose Al-F groups were slight. Conclusion: High dose Al could inhibit the growth of osteoblasts in culture in vitro and had certain extent toxicity to them. Fluoride could inhibit the cell toxicity of aluminum in certain extent. The effects of high dose Al and high dose Al-F on the ultrastructure of osteoblasts mainly showed the damage of the membrane structures.
