摘要
球形芽孢杆菌BS10(Bacillus sphaericus Strain 10,BS10)是从我国分离得到的一株高毒力杀蚊幼虫菌株。BS10在芽孢形成过程中产生的伴孢晶体蛋白对蚊幼虫,特别是库蚊幼虫具有强的毒杀作用。本文建构了BS10的重组克隆,并通过合成18碱基的寡聚核苷酸序列为探针,筛选出了BS10的重组阳性克隆[TGl(pFL37)和TGl(pFL36)]。重组克隆TGl(pFL37)含有编码43kd毒素蛋白的4.0kb Hind Ⅲ的BS10 DNA片段。Western blot分析和生物活性测定证明BS10的43kd杀蚊毒素蛋白基因在大肠杆菌中得到表达。
Bacillus sphaericus strain 10 (BS10), isolated from Jiang Su Province of China, is highly toxic to the mosquito larvae. During sporulation, BS10 produces a parasporal crystalline protein which is toxic to the larvae of a number of mosquito species, and extremol high toxic against the larvae of Culex pipiens. Using the Escherichis coli cloning vcctor pAT153, two clones positively hybridized with the synthesizing 18 bases oligonucletides probe have been obtaubcd. One of the recombinants, TG1(pFL37), contains a 4.0 kb Hind Ⅲ DNA fragment from BS10, and produces the 43 kb larvicidal toxin protein. Western blotting anal, sis and biological assay of mosquito-larvicidal activity confirm that larvicidal toxin gene of BS10 is expressed in E. coli.
