摘要 目的:建立以反相高效液相色谱法测定清肺合剂中东莨菪内酯含量的方法。方法:色谱柱为C18(250mm×4.6mm,5μm),流动相为甲醇-0.5%醋酸(40∶60),检测波长为345nm,流速为1.0mL·min-1。结果:东莨菪内酯进样量在0.04~0.80μg范围内与峰面积积分值呈良好线性关系(r=0.9999);平均回收率为98.6%,RSD=1.35%(n=6)。结论:本方法方便易行、结果准确、重复性好,可用于清肺合剂的质量控制。 OBJECTIVE: To establish an RP-HPLC method for the content determination of scopoletin in Qingfei mixture. METHODS:The C18(250mm×4.6mm,5μm)column was used with the mobile phase consisted of methanol-0.5% acetic acid (40: 60) at the flow rate of 1.0 mL·min^-1. The detection wavelength was set at 345 nm. RESULTS: It showed a good linear relation between scopoletin content and peak area at a range of 0.04-0.80 μg(r=0.999 9), and the average recovery rate was 98.6% (RSD= 1.35% ,n=6). CONCLUSION: The method is simple, accurate and highly reproducible for the quality control of Oin-fei mixture.
