场强放大样品进样-毛细管电泳技术对中药制剂中苯甲酸与山梨酸的测定 被引量:6

Determination of Benzoic Acid and Sorbic Acid in Peroral Preparations of Chinese Herbal Medicine by Field Amplified Sample Injection with Capillary Electrophoresis

摘要 利用场强放大样品进样一高效毛细管电泳分离技术测定中药口服制剂中苯甲酸和山梨酸的含量。采用未涂层熔融石英毛细管柱(50.2Cm×75μm,有效长度40cm),以pH9.5的50mmol/L硼砂为运行缓冲液,柱温25℃,分离电压20kV,PAD检测,检测波长228nm。阳极进样,进样前压力进水3.45kPa×5s,电动进样-10kV×3s。在最佳富集条件下,苯甲酸和山梨酸的富集倍数(以峰面积计)可达259.7和231.9倍;在0.5—20.0mg/L范围内呈良好的线性关系,相关系数高于0.995,检出限分别为0.67、0.53μg/L。将此方法用于两种液体中药制剂中苯甲酸和山梨酸的测定,苯甲酸的回收率为98%~103%,山梨酸的回收率为100%-101%。该法简单快速、准确可靠、灵敏度高、无需复杂仪器,为液体中药制剂中防腐剂的含量控制提供了新的检测方法。 A method of field amplified sample injection with capillary electrophoresis was developed for the analysis of benzoic acid and sorbic acid in peroral preparations of Chinese herbal medicine. The separation was achieved on a fused-silica capillary column (50. 2 cm × 75 μm, effective length: 40 cm) using pH 9.5 50 mmoL/L Na2 B4 07 as a running buffer with an applied voltage of 20 kV and a cartridge temperature of 25 ℃. The PAD detection wavelength was set at 228 nm, and 3.45 kPa x 5 s water plug was introduced from the anode before injection. Sample was injected with electrokinetic injection by - 10 kV x 3 s. The calibration curves of sorbic acid and benzoic acid were linear in the range of O. 5 - 20. 0 mg/L with correlation coefficients more than 0. 995. The limits of detection were 0. 67 ixg/L for benzoic acid and 0. 53 μg,/L for sorbic acid. The stacking multiples( counting by peak area) of benzoic acid and sorbic acid were 259.7 and 231.9, respectively. Under the optimal condition, the proposed method was applied in the determination of benzoic acid and borbic acid in the peroral preparations of Chinese herbal medicine with recoveries of 98% - 103% for benzoic acid and 100% -101% for sorbic acid. This method was simple, rapid, accurate and sensitive, and could be used to determine the content of preservatives in biological sample.
