摘要
本实验以大麦叶肉原生质体为材料,采用常规原生质体分离培养方法,在含有1.5mg/l2,4-D、0.5 mg/l 6-BA和0.2 mg/l IAA的D_(2α)培养基中诱导了原生质体持续分裂,形成了小愈伤组织,但它未能进一步发育分化产生绿苗。通过对原生质体分离、分裂影响因素的探讨,我们认为材料的生理状态以及遗传背景在培养中起了关键作用。
Large number of mesophyll protoplasts were isolated from barley leaves, subsequently sustained cell division was induced and microcalli formed when cultured on D_2a medium supplemented with 1.5mg/l 2,4-D, 0.5mg/1 6-BA and 0.2mg/1 IAA. But the microcalli could not develop further. After many attempts, we suggest that the critical factors seem to be associated with physiological state and genetic background of materials.
