摘要
目的探讨白介素27(IL-27)对哮喘患儿干扰素γ(IFN-γ)及白介素4(IL-4)的调节作用。方法收集23例哮喘患儿外周血单个核细胞(PBMC),加入不同浓度(5ng/mL或0.5ng/mL)重组人白介素27(rhIL-27)进行体外培养12h,予酶联免疫吸附法(ELISA)测定其培养上清IFN-γ及IL-4浓度。结果 5ng/mL和0.5ng/mL两个浓度的rhIL-27刺激PBMC进行体外培养后,IFN-γ表达分别为85.9±12.2和8.9±2.3μg/L,较细胞刺激素组(7.2±1.3μg/L)均有显著升高(P<0.01或0.05)。5ng/mLrhIL-27组IL-4表达较细胞刺激素组有明显降低(15.0±1.9μg/Lvs77.0±15.6μg/L,P<0.01)。结论 IL-27参与了哮喘的发病过程,它可通过调节Th1/Th2相关细胞因子活性为哮喘治疗提供一种新的途径。
Objective To study the effect of interleukin(IL)-27 on INF and IL-4 activities in children with asthma.Methods The levels of IFN-γ and IL-4 in culture supernatants of peripheral blood mononuclear cells(PBMCs) from 23 asthmatic children were measured using ELISA.PBMCs were cultured with recombinant IL-27(5 ng /mL or 0.5 ng /mL)for 12 hrs in vitro.Results IFN-γ levels in both 5 ng /mL and 0.5 ng /mL IL-27 treatment groups(85.9 ± 12.2 and 8.9 ± 2.3 μg /L,respectively) increased compared with those in the cytokine stimulated control group(P〈0.01,P〈0.05 respectively).The group treated with 5 ng /mL IL-27 had decreased significantly IL-4 levels compared with the cytokine stimulated control group(15.0 ± 1.9 μg /L vs 77.0 ± 15.6 μg /L;P〈0.01).Conclusions IL-27 may be involved in the pathogenesis of asthma.It may affect Th1 /Th2 cell activities and might be a new option for the treatment of asthma.
