摘要
目的探讨1L-27对支气管哮喘(简称哮喘)患者干扰素y(lFN-γ)及1L-33的调节作用。方法选择56例非细菌性感染所致中-重度哮喘患者为试验病例组,整群匹配方法选择30名正常者为对照组,酶联免疫吸附试验法检测1L-27、干扰素y(IFN-y)、1L-33水平,全血细胞分析仪检测白细胞数、中性校细胞绝对值、嗜酸粒细胞绝对值;同时收集哮喘组患者外周血单个核细胞(PBMC).加入不同浓度的(5μgiL或0.5μgiL)重组人白介素27(rh1L-27)进行体外培养12 h.酶联免疫吸附试验法检测其培养 上清1FN-y及1L-33浓度;探讨1L-27对1FN-γ、1L-33的影响及各指标间的相关性。结果哮喘组患者血清1L-33较对照组明显升高(t=34. 53. Objective To study the effect of interleukin (IL)-27 on INF-y and IL-33 activities in patients with bronchial asthma (asthma). Methods Two groups of experiments, the serum levels of IFN-y and IL-27 and IL-33 in asthmatic patients or control group by ELISA. The levels of WBC and neutrophils and EOS by hematology analyzer. PBMCs from 33 asthmatic patients were cultured with recombinant human IL-27 (5 μg/L or 0. 5 μg/L) for 12 hours in vitro, and then the levels of IFN-y and IL-33 in their supernatant were measured by ELISA. The explore of correlation between IL-27 activating factor and IL-33 , IFN-y in the asthmatic patients. Results The levels of IL-33 in asthmatic patients were significantly increased and IFN-y, IL-27 had a significant decrease compared with that in healthy control group (t IFN-y=27. 52, P IFN-y〈0. 01; t IL-27 =32.77, P IL-27 〈0. 01). The values of IFN-y in supernatant were (85.9 ± 12.1) flg/L, (19. 6 ± 3.2) μg/L compared with that in cytokine stimulated control group (11. 3±2. 7) μg/L had increased significantly ( t = 34.57, P 〈0. 01; t = 11. 39, P 〈0. 01). The group treated with 5 μg/L IL-27 had decreased significantly IL-33 levels compared with the cytokine stimulated control group [(33. 2± 11. 8) μg/L vs (116. 9± 17.6) μg/L, t = 22. 69, P 〈0.01]. There was a positive correlation between IFN-y and IL-27 in asthmatic patients. Conclusions The levels of IL-27 ,IFN-y, IL-33 were interelated.
