雷公藤多糖甙对大鼠脑缺血/再灌注损伤的影响 被引量:4

Protective effects of multi-glycosides of triptergium wilfordii Hook F. on cerebral ischemia-reperfusion injury in rats

摘要 目的:研究雷公藤多糖甙(GTW)对脑缺血/再灌注损伤的影响。方法:132只Wistar大鼠分为4组,每组33只:Ⅰ组为假手术组,Ⅱ组为手术组,Ⅲ组为生理盐水治疗组,Ⅳ组为GTW治疗组。Ⅳ组于实验前3d开始灌胃给30mg/kgGTW。Ⅲ组给予等体积生理盐水。各组大鼠分别于灌胃后1、3、6、12及24h时间点处死6只,留取血清用放射免疫法测定肿瘤坏死因子-α(TNF-α)含量。每组3只缺血1h/再灌注24h后,取脑制作病理组织切片观察各组脑组织形态变化。结果:Ⅳ组多数神经元结构完整的,形态相对正常,间质水肿轻。再灌注3、6、12及24h,Ⅳ组较Ⅱ、Ⅲ2组TNF-α含量降低(F=58.971,113.639,68.395和60.011,P<0.001)。结论:GTW对大鼠及局灶性脑缺血/再灌注损伤有保护作用。 Aim:To study the role of tumor necrosis factor α(TNF-α) on focal cerebral ischemia-reperfusion injury in rats and the effects of multi-glycosides of tripterygium wilfordii Hook F. (GTW) on them.Methods:Rat cerebral ischemia-reperfusion injury model was established with suture method.A totle of 132 Wistar rats were randomly divided into four groups and 33 in each group. Group Ⅰ was sham-operated,group Ⅱ was operation group,group Ⅲ was normal saline treated group,group Ⅳ was GTW treatment group.The rats of group Ⅳ were given GTW 3 d before the experiment. The rats of group Ⅲ were given the same doses of normal saline.Six rats in each group were sacrificed at affer 1,3,6,12,and 24 h,and TNF-α contents of each group in serum were measured by using radioimmunoassay method at each time.Three rats in each group were decapitated after ischemia 1 h/reperfusion 24 h and brain was removed to make pathological tissue slice.Results:The majority neurons of treatment group have integrated structure,relative normal shape and mild interstitial edema. At reperfusion 3,6,12,24 h,the TNF-α of group Ⅳ was less than groups Ⅱ and Ⅲ (F=58.971,113.639,68.395,60.011,P0.001),which showed that GWT could inhibit rise in TNF-α,and reduce the concentration of TNF-α.Conclusion:GTW possesses protective effect on focal cerebral ischemia-reperfusion injury in rats.
