摘要
目的研究大鼠脑缺血/再灌注时热休克蛋白(HSP)70表达及川芎嗪对HSP70表达的影响。方法采用SD大鼠左侧颈总动脉结扎并滴注生理盐水脑缺血/再灌注模型,以免疫细胞化学法检测24只缺血/再灌注及川芎嗪干预大鼠脑缺血/再灌注30min时脑组织HSP70表达并与病理组织学改变进行对照研究。结果(1)非缺血侧脑组织无HSP70表达,缺血30min缺血侧大脑皮层可见HSP70表达;再灌注30min组仅1只大鼠(1/6)缺血侧大脑皮层有HSP70表达。(2)与单纯缺血大鼠相比,川芎嗪干预组缺血侧大脑皮层HSP70表达明显增强,计算机辅助图像半定量分析HSP70免疫阳性细胞数,分别为平均43.55±12.51个/片和84.95±16.7个/片(P<0.01)。(3)缺血侧皮层神经细胞呈现轻度缺血改变,川芎嗪干预大鼠缺血损伤程度似有减轻。结论脑缺血时可诱导缺血脑细胞部分HSP70表达,川芎嗪干预可促使缺血大脑皮层HSP70表达明显增强,皮层神经细胞的缺血损伤有所减轻。
The expression of HSP 70 and the effects of tetramethylpyrazine(TMP) on cerebral ischemic reperfusion injury(CIRI) in rats were studied. Methods Using an experimental cerebral ischemia model in Sprague Dawley(SD) rats, the cerebral cellular expression of HSP 70 and the effects of TMP, a blood activating and statsis eliminating herb, on CIRI were studied by immunohistochemical staining in conjunction with histopathological analysis of rat brain. Results (1)No HSP 70 was found in none ischemia brain tissues and HSP 70 was observed in the parietal cortices 30 minutes after brain ischemia. But, 30 minutes after reperfusion, no HSP 70 was observed except in one rat out of the six. (2)Compared with that of cerebral ischemia rats, HSP 70 expression increased significantly and the number of HSP 70 immunoreaction positive cells in cerebral ischemia rats with TMP increased markedly by computer image analysis(from 43.55±12.51/section to 84.95±16.7/section,P<0 001).(3)After cerebral ischemia, most neurons were found morphologically intact except for some mild cerebral ischemic changes in the parietal cortex, such as neuron and nucleolus shrinkage and smeared nucleolar cytoplasmic border. In the TMP group, though some mildischemic changes in the parietal cortex. still exist, they were alleviated, with no neuron and only slight nucleolus shrinkage.Conclusion Cerebral ischemia could induce the HSP 70 protein in injured cells and the administration of TMP could increase HSP 70 expression and alleviate cerebral ischemic changes histopathologically.
