摘要
从新生仔猪颈部皮下采集脂肪组织,用胶原酶Ⅳ消化分离前脂肪细胞,于37℃,5%CO2、饱和湿度下培养前脂肪细胞,通过MTT法、油红O染色及脂蛋白脂酶(LPL)活性测定对培养细胞进行鉴定.结果表明:培养3 d后,细胞增殖进入对数增长期,其生长曲线呈S形;分化的脂肪细胞内充满脂肪滴,细胞形态由梭形逐渐变为圆形;随着前脂肪细胞的分化,LPL活性显著下降.
The adipose tissue collected from subcutaneous of newborn crossbred piglets,was digested by collagenase Ⅳ,then the porcine preadipocytes were isolated and cultured subsequently in 37℃,5%CO2 and saturation humidity.The proliferation and differentiation of the preadipocytes were recognized by MTT and Oil Red O staining method,and the lipoprotein lipase(LPL) activity of the cultured cells was also detected.The results showed that the amount of cultured cells tend to be exponentially increased 3 days later and the growth curve showed itself "S" figure.At the same time,the fatty droplets in differentiating adipocytes were observed.Henceforward,more differentiated cells containing fatty droplet or fusioned fatty dropelet were appeared,and the morphology of the cells also changed from fibroblast-like to round accordingly.Moreover,along with preadipocytes differentiation,the activity LPL of the cells decreased remarkably.The results obtained above establisha foundation for the further study of proliferation and differentiation mechanism of porcine preadipocytes.
