摘要
牛卵母细胞用 5 % CS的 TCM- 1 99培养液体外成熟培养 ( IVM) 2 2 h;以 BO液 +咖啡因作精子获能处理 1 .5 h;BO液 + BSA的培养液体外受精 ( IVF) 5 h;受精卵 (含卵丘细胞 )用 5 %CS的 CR1 aa培养液在 5 % CO2 、95 %空气、38.5℃、饱和湿度的培养环境中体外培养 ( IVC) 7~ 8d,结果受精卵裂率为 72 .4% ( 5 6.3%~ 81 .0 % ) ,囊胚率 2 3.7% ( 1 2 .5 %~ 40 .9% )。
Bovine oocytes(n=228) were incubated for 22 h in TCM 199 with 5% calf serum(CS); The sperm suspension was pre incubated for 1.5 h in Brackett and Oliphant medIUm(BO) with caffeine; The oocytes were cultured with sperm for 5 h in BO supplement with bovine serum albumin(BSA). Fertilized oocytes were cultured for days 7~8 d in CR1aa with 5% CS. All incubations were conducted at 38.5℃ in 5% CO 2, 95% air and in the saturated humidity. The results showed that the rates of cleavage and blastocyst of fertilized oocytes were 72.2%(56.3% to 81.0%) and 23.7%(12.5% to 40.9%), respectively.
