摘要
目的:探索葛根素(puerarin)对大鼠光损伤视网膜中NF-κB表达的影响。方法:48只SD大鼠随机分为空白组、模型组、低剂量组和高剂量组。采用YZ20T眼科手术显微镜,距角膜距离为(150±3)mm照射右眼,照射时间为30 min,照度为(22 000±1 000)勒克斯建立急性光损伤模型。低剂量组、高剂量组尾静脉注射葛根素,空白组、模型组尾静脉注射等量生理盐水。每日1次,连续给药7 d。7 d后取眼球,观察视网膜外核层组织形态学及检测外核层细胞数,通过免疫组化测定视网膜中NF-κB的表达。结果:模型组视网膜外核层变薄,细胞减少,细胞排列较正常组稀疏,内外节排列紊乱,分界不清;低剂量组视网膜形态有改善。空白组视网膜外核层细胞浆中NF-κB阳性物质含量较多,模型组视网膜外核层细胞浆中NF-κB阳性物质含量较少。各用药组视网膜外核层细胞浆中NF-κB阳性物质有不同程度增加。结论:葛根素对手术显微镜导致的视网膜光损伤有修复作用,其作用机制可能是通过增加NF-κB的表达,从而拮抗细胞凋亡。
Objective:To observe effects of puerarin injection on NF-κB expression in retina photic injury rats induced by operation microscope.Methods:48 SD rats were randomly divided into blank control group,model group,puerarin injec-tion treatment group 1(low dosage)and group 2(high dosage).Ratina photic injury models were induced by operation mi-croscope.Histomorphology changes of outer nuclear layer of retina were observed,the changes of NF-κB expression in retina were detected by immunohistochemistry.Results:Compared with model group,histomorphology was improved in treatment group 1;expression of NF-κB in model group is lower than that in blank control group.Compared with model group,expression of NF-κB was significantly increased in treatment group 1.Conclusion:Puerarin is likely to repair reti-na photic injury by upregulating NF-κB expression to against apoptosis in rats.
