摘要
目的观察滋阴明目丸对视网膜光损伤大鼠的保护作用及机理。方法将70只Sprague-Dawley(SD)大鼠随机分为7组,除空白对照组外均采用自制光损伤装置造成视网膜光损伤模型,光照后3天开始灌服滋阴明目丸及对照药物,连续15天,取大鼠视网膜检测基因p53、Bcl-2的表达。结果滋阴明目丸可显著降低视网膜p53的表达,增强Bcl-2的表达,与模型对照组比较差异有显著性(P<0.01)。结论滋阴明目丸可通过影响凋亡相关基因p53、Bcl-2的表达,从而抑制视细胞的凋亡。
OBJECTIVE To investigate the protective effect and mechanism of ziyinmingmuwan (滋阴明目丸) on retina against photic injury. METHODS Seventy Sprague - Dawley (SD) rats were randomly divided into 7 groups, with 6 groups prepared as the models of photic trauma at first and another group taken as blank controlling one among these groups. After three days, the animals in the treatment and controlling groups were treated with ziyinmingmuwan and control agent respectively for 15 days during the period of experiment, following this, the retinal tissues were taken to test the expressive activities of p53 and Bcl - 2 genes by immunohistochemical method. RESULTS Compared with the model group, the expression of p53 was decreased and Bcl - 2 was increased evidently in groups of ziyinmingmuwan, which showed significant difference ( P 〈 0.01 ) .CONCLUSIONS Ziyinmingmuwan could inhibit the expression of p53 gene and enhance the expression of bcl - 2 gene to induce an inhibitory effect on apoptosis of the retinal cells.
