摘要
目的通过比较建模成功率,找到建立大鼠海人酸(kainic acid,KA)颞叶癫痫模型的合适用药剂量。方法雄性SD大鼠80只,体重240-260g,随机分为4组,每组20只,分别向右侧海马注射KA1.0μl,0.7μl,0.4μl和生理盐水1.0μl,KA浓度0.5μg/μl,术后观察4h,按照Racine的癫痫大鼠发作时的行为学分级标准,连续3次达到Ⅳ-Ⅴ级发作者确定成功模型。结果观察结束时,生理盐水组大鼠全部存活且未见痫性发作。KA1.0μl组死亡14只,Ⅴ级发作6只。KA0.7μl组死亡2只,Ⅴ级发作15只,Ⅳ级发作3只。KA0.4μl组死亡1只,Ⅴ级发作1只,Ⅳ级发作6只,Ⅲ级发作9只,Ⅱ级发作3只。经统计学检验,KA0.7μl组大鼠颞叶癫痫模型的成功率最高。结论 0.7μlKA右侧海马注射较易获得大鼠颞叶癫痫模型。
Objective To explore a proper dose of kainic acid (KA) inducing epilepsy model in rats. Methods Eighty male SD rats,weighing 240-260 g,were randomly divided into 4 groups(n=20 in each group).Then the kainic acid at the concentration of 0.5 μg/μl for 1.0 μl,0.7 μl,0.4 μl and saline 1.0 μl were respectively injected into the right hippocampus of rats in 4 groups.After the injection,the behaviors of all rats were recorded for 4 h according to Racine's classification standard (1972).The rat with 3 consecutive seizures of Grade Ⅳ-Ⅴ was defined as a successful model of epilepsy. Results In saline group,all rats survived,and no seizure was observed.In KA 1.0 μl group,14 rats died,and 6 rats reached grade Ⅴ.In KA 0.7 μl group,2 rats died,15 rats reached grade Ⅴ,and 3 rats reached grade Ⅳ.In KA 0.4 μl group,1 rat died,1 rat reached grade Ⅴ,6 rats reached grade Ⅳ,9 rats reached grade Ⅲ and 3 rats reached grade Ⅱ before the end of the observation. Conclusion KA 0.7 μl has the best effect for establishing the model of epilepsy.
