丘脑前核电刺激对癫痫大鼠海马细胞外谷氨酸和γ-氨基丁酸含量的影响 被引量:2

Changes of glutamate and y- aminobutyric acid of extracellular fluid in hippocampus during electrical stimulation of anterior nucleus thalamus in rats

摘要 目的通过电刺激癫痫模型大鼠双侧丘脑前核,观察海马细胞外液中谷氨酸、γ-氨基丁酸(GABA)含量的变化,探讨双侧丘脑前核电刺激治疗癫痫的机制。方法应用立体定向技术建立大鼠海人酸癫痫模型,应用刺激电极对双侧丘脑前核进行电刺激,同时收集海马细胞外液,用高压液相色谱法检测细胞外液谷氨酸和GABA的含量。结果癫痫大鼠海马细胞外谷氨酸[(2.10±1.12)μmoL/L]明显高于对照组大鼠[(1.05±0.38)μmoL/L],而GABA的含量[(0.28±0.21)μmoL/L]低于对照组大鼠[(0.75±0.27)μmoL/L]。癫痫组大鼠在电刺激后,海马细胞外液中谷氨酸明显减少,GABA明显增加。而对照组大鼠海马细胞外谷氨酸和GABA在刺激前后未见明显变化。结论海马细胞外液谷氨酸下降及GABA升高在丘脑前核电刺激治疗中起重要作用。 Objective To explore the changes of extracellular glutamate (Glu) and y-aminobutyric acid (GABA) in hippocampus during the high-frequency stimulation of anterior nucleus thalamus (ANT) in epileptic rats. Methods A total of 30 rats were divided equally into 2 groups : epilepsy model and normal control (n = 15 each). Concentric bipolar electrodes were stereotaxically implanted in bilateral ANT. A high- frequency stimulation of 130 Hz was applied in the epilepsy group. Mierodialysis probes were unilaterally lowered into hippocampus. The concentrations of Glu and GABA in dialysate samples were determined by high-performance liquid chromatography ( HPLC ) . Results Electrical stimulation of ANT induced an increase of GABA and a decrease of Glu in hippocampus. Condusion An increase of GABA and a decrease of Glu durin~ the electrical stimulation of ANT nlav an imoortant role in the theraDv of enilensv.
