微生物法拆分制备S-倍他洛尔中间体

摘要 对利用胶红酵母DQ832198催化倍他洛尔中间体拆分的转化条件进行优化.每次只优化一个条件,其他条件固定为已选定的结果.优选出的最佳转化条件为:菌体培养时间16 h,转化温度28℃,在pH 6的Na2HPO4-柠檬酸体系中转化,底物加入量3 g/L,豆浆、番茄汁和K+对拆分有利.转化中的关键立体选择性酶为操作稳定性良好的非胞外脂肪酶.利用优化后的条件进行转化,转化速度有所提高. The stero-resolution conditions of betaxolol intermediate by Rhodotorula mucilaginosa DQ832198 are optimized. Only one conversion condition is optimized everytime, the others kept unchanged as already chosen. The optimum raising period is 16 h, the optimum conversion temperature is 28℃, the most suitable conversion pH is 6 in the Na2HPO4-citric acid system, and the optimum substrate concentration is 3 g/L. Soybean milk, tomato juice and K+ have advantageous effect on bioconversion. The crucial stereo-selectiving enzyme in the conversion is a non-endocellulous lipase with excellent stability. Optimization of the conversion conditions accelerates the conversion course.
