摘要
目的观察猪胎盘替代紫河车用于防治老年性痴呆的效果。方法将70只昆明种小鼠分为空白组、模型组、阳性对照组、紫河车低剂量组、紫河车高剂量组、猪胎盘低剂量组、猪胎盘高剂量组,每组10只。以D-半乳糖皮下注射法制备老年痴呆动物模型,空白组不给任何药物,模型组以生理盐水代替受试药,阳性对照组灌胃给予脑复康,紫河车低剂量组、高剂量组分别灌胃给予紫河车28g/kg、4g/kg,猪胎盘低剂量组、高剂量组分别灌胃给予猪胎盘4g/kg、8g/kg。给药6周后,分别运用行为学实验避暗法、跳台法,测定两药对小鼠造模后学习记忆的影响,并测定脑中乙酰胆碱酯酶和单胺类神经递质的含量。结果猪胎盘和紫河车抗老年痴呆作用无统计学差异(P<0.05),在避暗潜伏期试验中,猪胎盘高剂量组的效果明显好于紫河车高剂量组(P<0.01)。两者对单胺类神经递质的影响相近。结论猪胎盘有可能替代紫河车用于防治老年痴呆。
Objective To observe the effect of pigplacenta instead of Ziheche （Placenta Hominis） for the treatment of senile dementia. Methods Seventy Kunming mice were divided into blank group, model group, positive control group, Ziheche low and high dosage groups, and pigplacenta low and high dosage groups, with 10 in each. The senile dementia models were established with the DGalactose subcutaneous injection. The blank group was not administered any medicines. The model group was prescribed normal saline instead of the tested medicine; the positive control group was given Naofukang by garage; the Ziheehe low and high dosage groups were given 2g/kg and 4g/kg Ziheche respectively by garage; while the pigplaeenta low and high dosage groups were treated with pigplacenta 4g/kg and 8g/kg respectively by gavage. After treatment for 6 weeks, the behavior experimental dark-avoiding test and stepdown test were applied to test the effect of the medicines on the learning memory of mice, and acetylcholinesterase and monoamine levels in brain tissues. Results There was no significant difference between the effect of pigplacenta and Ziheche in resisting senile dementia. In the latency of dark-avoiding test, the effect of high dosage of pigplacenta was significantly better than that of Ziheche （P〈 0.01）. The effect of pigplacenta and Ziheche on monoamine was similar with each other. Conclusion It＇s possible to use pigplacenta instead of Ziheche for the treatment of senile dementia.
