摘要
本文对光合细菌的培养条件进行研究后,发现适当增加菌种的接种量和光照强度、以及微好氧培养,均会提高光合细菌的生长速度。采用石油醚──甲醇抽提法对菌体中的类胡萝卜素进行提取,最终获得的类胡萝卜素产量为170mg/L培养液,菌体湿重为7.5g/L培养液。对所获得的色素的部分性质进行分析,通过薄层层析发现其中含有三种组分。该色素对酸较为稳定,对热的稳定性则较差。在室内无线条件下,具有一定的稳定性,可以较长时间地保存,但在室外阳光直射处便会迅速分解。
The extraction technology of carotenoid from photosynthetic bacteria was studied. The bacterial cells were a ccumulated with a suitable medium. The growth could be accelerated when the inoculative volume, illumination intensity or ventilation was enhanced properly. By means of petroleum ether-methanol, carotenoid was extracted from the cells. The yield of carotenoid in ferment medium was 170mg/L,and the wet weight of the cells was 7.5g/L. The properties of carotenoid were studied, and three kinds of components were found by thin-layer chromatography.Carotenoid was stable against acid, but not against heat. It could be preserved for a long time in indoor rays, but decomposed quickly in sunlight.
