摘要
试验采用响应面方法优化光合细菌Rhodospirillum Rubrum S1培养基。结果表明:培养基成分中柠檬酸、牛肉膏、MgSO4和FeSO4对于光合细菌的生长影响最为显著。最优培养基配方:柠檬酸为3.5291g/L、牛肉膏为3.3190g/L、MgSO4为0.5083g/L、FeSO4为0.0194g/L。此时最大响应值为13.77mg/L。验证实验结果表明,采用优化后的培养基,光合细菌的类胡萝卜素产量达到13.69mg/L,与理论值相差0.58%。因此,利用响应面法对光合细菌的培养基进行优化合理可行。
Response surface methodology is used to optimize the culture medium for photosynthetic bacteria (PSB) Rhodospirillum Rubrum S1. The result shows that citric add, beef extract, MgSO4 and FeSO4 have the most significant influence on the growth of Rhodospirillum Rubrum S1. The optimized medium is as follows: citric acid of 3. 5291 g/L, beef extract of 3. 3190 g/L, MgSO, of 0. 5083 g/L, FeSO4 of 0. 0194 g/L, the maximum response value now is 13.77 mg/L. Then veri[ieation experiment is carried out, which indicates that with the optimized culture medium, the output of earotenoids rise to 13. 69 nag/ L, 0. 58% less than its theoretical value. Therefore, the expolitation of response surfaee methodology in optimizing PSB Rhodospirillum Rubrum S1 is reasonable and feasible.
