左归丸对阴虚阳亢大鼠Th1/Th2细胞漂移特异性转录因子T-bet及GATA-3 mRNA的干预作用 被引量:5

摘要 目的:观察阴虚阳亢大鼠Th1/Th2细胞漂移现象、两类细胞特异性转录因子T-bet及GATA-3 mRNA地表达及左归丸对阴虚阳亢大鼠Th1/Th2细胞漂移特异性转录因子T-bet及GATA-3 mRNA的干预作用。方法:30只大鼠随机分成空白对照组(A组)、阴虚阳亢组(B组)、左归丸组(C组),B、C两组采取腺嘌呤法建立阴虚阳亢模型,然后C组用左归丸滤液灌胃,A、B两组大鼠用生理盐水灌胃,RT-PCR法分别检测用药后大鼠脾脏T-bet及GATA-3 mRNA的表达。结果:B、C组造模后、用药前T-bet mRNA表达明显上调,而GATA-3 mRNA表达下调,经左归丸组干预后,T-bet mRNA表达明显下调,而GATA-3 mRNA表达上调。结论:左归丸干预Th1/Th2细胞漂移的途径可能是通过下调Th1细胞特异性转录因子T-bet mRNA表达,上调Th2细胞特异性转录因子GATA-3 mRNA表达,使阴虚阳亢大鼠Th1/Th2细胞达到一种新的平衡。 Objective: To explore the intervention Of Zuogui pill on specific transcription factor T-bet and GATA-3 in rats with hyperactivity of Yang due to yin deficiency and Th1/Th2 cell shift and intervention of Zuogui pill on specific transcription factors - T-bet and GATA-3. Methods: 30 rats were randomly divided into 3 groups, controlled group (Group A), group with hyperactivity of yang due to Yin deficiency ( Group B) and Zuogui pill group (Group C), The Models of hyperactivity of yang due to Yin deficiency were made in Group B and C, Rats in Group C were fed with filtrate of Zuogui pill, however, the rats in Group A and B were fed with physical saline solution, the expression of T-bet and GATA-3 mRNA were determined after the administration of filtrate of Zuogui pill. Results: the expression of T-bet mRNA in group B and C were raised before the administration of physical solution, but the expression of GATA-3 were decreased, however, after the intervention of Zuogui pill, the expression of GATA-3 were raised, the expression of T-bet were decreased. Condusion: Zuogui pill may interfere Th1/Th2 cell shift through down-regulating expression of T-bet mRNA and up-regulating expression of GATA-3 mRNA, and reaching a new balance of Th1/Th2 cell.
