摘要
α-淀粉酶活性的调控是小麦生产和加工的一个重要问题。本研究比较了大麦、小麦、黑麦和小黑麦α-淀粉酶抑制蛋白对小麦α-淀粉酶作用的等电聚焦电泳图谱,并测定了具有大麦α-淀粉酶抑制基因或小麦种子休眠基因的发育种子内源α-淀粉酶同工酶谱。结果表明,大麦α-淀粉酶抑制蛋白可在小麦遗传背景中抑制α-淀粉酶-1活性。小麦种子萌动时,α-淀粉酶抑制蛋白使α-淀粉酶-1受到抑制,α-淀粉酶-2的合成以及整体整个发芽过程也随之推迟。在具有大麦α-淀粉酶抑制基因或小麦种子休眠基因的小麦发育种子中,α-淀粉酶-1活性均可较早消失,但受不同遗传背景影响。这些结果说明大麦α-淀粉酶抑制基因在小麦遗传背景中表达良好,可用于控制小麦种子穗发芽。
The control of α-amylase activity is of great significance in wheat production
and processing. The effects of α-amylase inhibitors from barley, wheat, rye and triticale on α-
amylase in wheat were studied by isoelectric focusing. The results showed that the barley α-amy
lase inhibitor gene in wheat genetical background could inhibit the activity of wheat a-amylase-1.
In the beginning of seed germination, barley α-amylase inhibitor inactivated α-amylase-1 in
wheat, and the synthesis of α-amylase-2 and total procedure of germination were postponed,
consequently. By α-amylase isozymogram analysis, it was found that α-amylase-1 activity termi
nated before seed maturity in the developing wheat seeds with barley α-amylase inhibitor gene or
wheat seed dormancy gene. The genetic background also affected the duration of α-amylase-1 ac
tivity. It was concluded that barley α-amylase inhibitor gene was well expressed in wheat genetic
background and could be used for controlling preharvest sprouting in wheat.
