摘要
目的:探讨腺样囊性癌(adenoid cystic carcinoma,ACC)组织中肝素酶(heparanase,HPA)和神经生长因子(nerve growth factor,NGF)及其受体P75的表达和相互作用,以及与ACC嗜神经侵袭之间的关系。方法:应用免疫组化SP法检测42例ACC组织中HPA、NGF和P75的表达,并对他们在不同病理类型和组织学部位的表达进行统计学分析。结果:NGF和P75在嗜神经(PNI)组和非嗜神经(NPNI)组的表达有统计学差异(P<0.05),两者呈正相关(r=0.429,P<0.05)。嗜神经组HPA和P75的表达正相关(r=0.558,P<0.05)。P75在神经周组织和远离神经部位的表达有显著性差异(P<0.05),NGF在神经组织和远离神经处的表达无明显差异。结论:在ACC的神经浸润中,NGF及其受体P75扮演着重要角色,但并非唯一因素,NGF可以通过和受体p75结合可以提高HPA的表达率和生物活性,进而促进ACC对神经组织的浸润。
Objective: To investigate mechanism of interaction of Nerve Growth Factor (NGF), its Receptor p75 and heparanase(HPA), as well as the relationships between adenoid cystic carcinoma(ACC) and perineural invasion by detecting their expressions in ACC, Methods: The expressions of NGF, p75 and HPA were detected by SP im munohistochemistry methods, and their expression levels in different pathological types and histological regions were analyzed. Results: The difference of positive expression of NGF and P75 between perineural invasion (PNI) group and non--perineural invasion (NPNI) group was significant(P〈0.05). The positive expressions of p75 and HPA were closely correlated PNI group(r=0. 558, P〈0.05). The expression levels of p75 in fasciculus had significant deviation compared with that in distant tumor tissues(P〈0.05). Conclusion: NGF and P75 played important roles in the PNI of ACC, but not as the only factors. The combination of NGF and p75 could enhance the expression and bioaetivity of HPA, which would open up more space for tumor cells and enhance the invasion into nerve tis sues.
