摘要
通过建立人肝细胞和肝窦内皮细胞原代分离和培养的方法,对严重烧伤后早期粘附分子在肝脏中的表达进行了探讨。从创伤意外死亡病人获取肝组织,根据库普弗细胞和内皮细胞贴附时间的不同,分离出肝窦内皮细胞和肝细胞进行培养;应用免疫组化方法对肝窦内皮细胞进行了鉴定;并用流式细胞仪动态观察烧伤病人(烧伤面积50%以上,Ⅱ~Ⅲ度,48h内)血清对两种细胞表达P-选择素和细胞间粘附分子-1(ICAM-1)的影响。结果显示,肝细胞正常情况下无P-选择素和ICAM-1表达,烧伤血清作用后从1h开始ICAM-1表达增强,6h达到一个高水平,后仍维持高水平;肝窦内皮细胞在正常情况下有少量P-选择素和ICAM-1表达,烧伤血清作用后P-选择素1h开始即有表达增强,2h有一降低期,6h达到一个高水平,后仍维持。ICAM-1的表达从2h开始即有明显增强,6h达到一个高水平,后仍维持。说明:①肝细胞正常情况下无P-选择素和ICAM-1表达,烧伤血清作用后ICAM-1表达增强;②肝窦内皮细胞在正常情况下有少量P-选择素和ICAM-1表达,烧伤血清作用后P-选择素和ICAM-1表达增强;③严重烧伤后早期中性粒细胞参与对肝细胞的损伤,粘附分子起着非常?
The expression of P selectin and ICAM 1 induced by burn patient's serum in the human hepatocytes and heptic sinosoidal endothelial cells(HSECs) were observed in this study. The human hepatocytes and HSECs were isolated and cultured, the flow cytometry was used to determine dynamically changes in the expression of P selectin and ICAM 1 on the hepatocytes and HSECs in response to burnt patient's serum. With burnt patient's serum, the expression of P selectin increased signifcantly at 1 hour(Event %:from 7.2 to 18.4; MFI: From 34.1 to 83.2; both P <0.01) in HSECs but not in hepatocytes, the expression of ICAM 1 increased significantly at 1 hour and 2 hours ( P <0.01) in hepatocytes(Event %: from 2.3 to 35.5; MFI: from 13.3 to 142.6) and HSECs(Event %: from 24.3 to 40.6; MFI: from 102.3 to 169.4). The expression of P selectin and ICAM 1 on the hepatocytes and HSECs play important roles in the aggregation of neutrophils in liver after severe burns during early phase, thereby leading to neutrophil mediated hepatocellular injury.
