摘要
采用体外内皮细胞培养,将一次性大面积切痂前后烧伤血清与内皮细胞孵育24h,观察内皮细胞病理形态改变及培养液中乳酸脱氢酶(LDH)、血管紧张素转换酶(ACE)活性和6-酮-前列腺素F1α(6-keto-PGF1α)含量变化,了解一次性大面积切痂前后烧伤血清对内皮细胞的影响。结果显示,一次性大面积切痂术前1d烧伤血清可使体外培养的内皮细胞脱落,细胞变形,细胞收缩间隙增大,胞浆出现空泡,核固缩。而术后1、3、7、14d烧伤血清对内皮细胞形态改变明显减轻;培养液中LDH、ACE活性和6-keto-PGF1α含量也均较术前烧伤血清组明显降低(P<0.01)。提示一次性大面积切痂能有效地减轻烧伤血清对内皮细胞毒性作用,从而在烧伤后多脏器功能障碍的防治中起重要作用。
The purpose of this study is to investigate the endothelial cell(EC) damage induced in vitro by burn serum which was obtained from patient before and after en masse escharectomy.Methods:Burn serum were obtained from patient 1d before and 1d,3d,7d,14d after operation.After incubating human umbilical vein endothelial cells with burn serum for 24 hours,morphological change was observed,and the levels of lactate dehydrogenase (LDH) and 6 keto PGF 1α and angiotensin converting enzyme(ACE) in medium were determined.Results:After being incubated with burn serum which obtained from patient on 1d preoperation,the HUVECs were deformed showing shrinkage of the cells and formation of intercellular space,and small vacuoles appeared in cytoplasm with karyopyknosis.Some of the ECs were detached.The level of LDH,ACE and 6 keto PGF 1α were high.However,when HUVECs incubated with burn serum isolated from patients on 1d,3d,7d,14d after operation,the ECs damage was minimized,and the activities of LDH and ACE and the content of 6 keto PGF 1α in medium were decreased significantly( P <0 01).Conclusion:Escharectomy en masse at one sitting could attenuate EC damage induced by burn serum.
