摘要
目的:观察兔颈动脉损伤后炎性因子TXB2及COX-2的动态变化,探讨动脉血管急性炎症用小檗碱干预的效果和其可能的机制。方法:30只日本大耳白兔随机分成假手术组(n=6)、模型组(n=6)、小檗碱组(n=9)、辛伐他汀组(n=9)。行兔颈动脉球囊扩张制备兔颈动脉损伤急性炎症模型术,于术后第3、6、10天自耳缘静脉取血4ml,分离血清及血浆,用放射免疫法测定血浆TXB2,酶联免疫吸附法测定COX-2。结果:(1)术后3天,假手术组、小檗碱组、辛伐他汀组COX-2浓度低于模型组(P<0.05),术后6天小檗碱组、辛伐他汀组低于模型组(P<0.05):(2)术后3天,假手术组、小檗碱组、辛伐他汀组TXB2浓度低于模型组(P<0.01),术后6天假手术组、小檗碱组、辛伐他汀组低于模型组(P<0.05)。结论:小檗碱对兔颈动脉损伤后急性期血栓素-B2等炎症因子有抑制作用,对血管急性炎症反应有应用价值。
Objective: To investigate the affect of Berberine on the expression of TXB2 and COX-2 after ballon injury of carotid artery in the rabbits.M.ethods:30 rabbits were randomly divided into four groups,control group(n=6),model group(n=6),befberine group(n=9)and simvastatin group(n=9).The control group tmtreated,the other three groups were treated with NS 5ml/(kg.d).Berberine 2.5mg/(kg.d) and simvastatin 3.3mg/(kg.d) resperatorly after carotid artery ballon injury .The blood sampale of serm drawn from vein 4ml from each one rabbit m 3.6.10 days after ballon injury.Radioimmunoassay was used to detect serm levels of TXB2. and the concentration of COX-2 by ELISA. Results:(t) the plasma levels of COX-2 in model group significantly higher than control group,berberine group and simvastatin group (P〈0.05) in 3 days after ballon injury,it was significantly higher than berberine group and simvastatin group (P〈0.05) in 6 days. (2)The TXB2 levels in contral group,berberine group and simvastatin group wa significantly lower than model group (P〈0.05),it was significantly higher than berberine group,simvastatin group and eontral group (P〈0.05) in 6 days. Conclusion:The Berberine had an Inhibition influence on the the serum or plasma levels of COX-2 nod TXB2 after rabbit carotid artery ballon injury.
